Solubility Measurements
Our solubility measurement service workflow typically uses the Echo Acoustic robot to directly transfer stock solutions using 10 mM DMSO stock solution. We then add the target solvent and incubate for 3 hours in a plate shaker before we screen the well-plates with Oryl’s proprietary instrument.
We deliver two types of graph per solubility measurement, one using second harmonic scattering and second using linear light scattering. Both measurements are acquired simultaneously and provide orthogonal and complementary results to strengthen data reliability. Once the measurements are done, our instrument automatically process the measured data, fit the results and generates the report for you. You will be able to explore the data in our data portal and perform further analysis.
We perform measurements in 384 well-plate at standard volume of 100 µL, 8-12 concentrations (2-fold serial dilution) per solubility datapoint with at least 3 replicates.
Depending on your target concentration, we can modify the concentration range and input this in our workflow. Compound dispensing is done with the Echo acoustic robot, so we ensure accurate dispensing. If needed, we can decrease the volume per well to <30 uL for limited API quantities.
