How the Oryl Solubility Measurement Service works

1. Experiment Setup

Define the type of measurements you need per API, per solvent (pH, ionic strength, excipients) and number of replicates. We will automatically acknowledge your order and provide you with estimate of costs and date of delivery of results. We will confirm your order as soon as possible.

2. Send Chemicals*

We perform test in 384 well-plates and we typically need ~2 μL of 10 mM DMSO stock per solubility datapoint. If you want to minimize compound consumption, we can perform low volume experiment (<30 uL per well). To reduce cost, we recommend that you send your APIs together with the solvents

3. Solubility/Aggregation Test

Oryl’s instrument is designed for high-throughput measurements with excellent sensitivity. Typically, we will deliver results within two weeks after we received your samples. Our instrument provides two orthogonal measurements: our proprietary second harmonic scattering together with ultra-sensitive linear scattering.

4. Report & feedback

Our instrument automatically process and fit your data and generate comprehensive reports. We will handle data analysis and provide a summary of the experiments.

*Before sending your chemicals, you need to agree to Oryl’s General Terms & Conditions (GT&C) and if needed, Oryl will sign your company’s T&C for material transfer agreement.

Solubility Measurements

Our solubility measurement service workflow typically uses the Echo Acoustic robot to directly transfer stock solutions using 10 mM DMSO stock solution. We then add the target solvent and incubate for 3 hours in a plate shaker before we screen the well-plates with Oryl’s proprietary instrument.

We deliver two types of graph per solubility measurement, one using second harmonic scattering and second using linear light scattering. Both measurements are acquired simultaneously and provide orthogonal and complementary results to strengthen data reliability. Once the measurements are done, our instrument automatically process the measured data, fit the results and generates the report for you. You will be able to explore the data in our data portal and perform further analysis.

We perform measurements in 384 well-plate at standard volume of 100 µL, 8-12 concentrations (2-fold serial dilution) per solubility datapoint with at least 3 replicates.

Depending on your target concentration, we can modify the concentration range and input this in our workflow. Compound dispensing is done with the Echo acoustic robot, so we ensure accurate dispensing. If needed, we can decrease the volume per well to <30 uL for limited API quantities.

Please indicate in your request if this is required. The plate layout below shows an example of a plate layout for 4 compounds with 3 replicates and two solvents, including positive and negative controls.

Compound Library Screening (DMSO)

We can perform non-destructive quality control (YES/NO, presence or absence of aggregates) of your compound libraries. Simply ship to us the well-plates where you store your compound libraries, properly sealed. We will test them in our instrument, provide you with a report and ship the well-plates back to you. You will get a YES (green) / NO (red) aggregation characterization for each compound.
